If you can not make any plays you can click on the next card from the reserves to set it atop the waste pile.

If you lose a level and click the end game button it is game over and you start at the beginning. If you refresh your web browser instead of clicking the game over button you can continue your prior game starting on the level you were at. Each card you remove from the tableau is worth a multiple of 50 points.
